Application of Recombinase-Mediated Nucleic Acid Amplification Technology for Detecting of Vibrio parahaemolyticus in Aquatic Products

Abstract: Vibrio parahaemolyticus is the main pathogen causing foodborneilness,and itisof great significance to establish arapid and accurate detection technology to prevent food poisoning causedby Vibrio parahaemolyticus. In this study,a highly effcient detection method for Vibrio parahaemolyticus was established basedonrecombinaseaided amplification (RAA).By comparing the sensitivity and specificity of the traditional culture method with the RAA technique,and optimizing the experimental parameters,the applicability of the RAA method in the detection of Vibrio parahaemolyticus was verified.The results show that RAA technology can directly detect the sample,and the results can be obtained in only 2h to 3h .For the low bacterial volume samples, the total detection time could be controlled within 10h after 6h to 8h of bacterial enhancement pretreatment, which significantly shortened the detection time. The established RAA detection method has high specificity ( 100% )and sensitivity,and is suitable for the on-site rapid detection of Vibrio parahaemolyticus in aquatic products and other foods.
