Comparative Study on Different Pretreatment Methods for Determination of Zearalenone in Grain by HPLC

Abstract: The effcts of different pretreatment methods on the analysis of zearalenone (ZEN) in grain were explored.In this paper,the effects of three extraction methods (oscilation,vortex,ultrasonic treatment) and two purification methods (to concentrate the eluent by nitrogen blowing or not)on the analysis of ZEN by high performance liquid chromatography (HPLC) were investigated by recovery and standard substances detection, and compared with the national standard method.The results indicated that the three extraction methods and two purification methods for pretreatment of grain samples could met the detection requirements. Considering improving the work efficiency,reducing operation process and saving the cost, the optimum combination of treatment methods was obtained that the extraction method was vortexing for 5min and ultrasonic treatment for 20min ,and the purification method was diluting the eluent directly with water.
