Determination of Total Alkaloids in Mulberry Branch by Pre-column Derivatization and HPLC

Abstract: Objective: To establish a pre-column derivatization and high performance liquid phase method for the determinationof totalalkaloids in mulberry plants.Methods: Samples werecolected with water and injected with 9-chloroformate methyl fluorene derivative reagent by HPLC with the volume of 20μL . The chromatographic analysis was performed at with red spectrum RDC18 5μm 4.6x250mm ) on the column and gradient elution of (citric acid buffer pH4.2 )(A)-acetonitrile (B) B at 0~22min , 47.5% Bat , 37.5% B→37.5% Bat 22.01~38min 37.5% B→75% Bat 75% Bat ). Theflow rate was 1.5 . Results: In the linear range of , the linear relationship was good , and the detection limit was . Conclusion: The method is rapid, simple and sensitive, and can be used for the determination of total alkaloid content in mulberry branch.
