Extractionand Antioxidant Activity of Total Flavonoids from Sea-buckthorn Branches and Stems

AbstractUsingsea-bucktobrancesasrmaterials,teetractionprocessoftotalfavonoidsasotiizedusinsinglefactorprmentsandresposesfaceethdology,nditsprotetieectolohlicliverjury(A)wssudiedTeesultssowdatetotal flavonoid content in the branches and trunks of sea-buckthorn was 0.64% . The optimal extraction process for total flavonoids from sea-buckthorn branches was ultrasonic frequency of 50kHz ,powerof 2OoW,extraction temperatureof 55°C ,liquid-material ratio of 23:1,ethanol concentration of 61% ,extraction time of 2 hours.Under these conditions,the total flavonoid extraction amount was 4 .19mg/g . Total flavonoids were purified and enriched using X-5 macroporous resin. In vitro antioxidant tests showed that the 40% ethanol purified product had the best antioxidantactivity.ThescavengingratesofDPPH,ABTS,hydroxylandsuperoxideanionradicalswere3.17,37.O6,792.51and (204號(hào) 262.42μg/mL ,respectively.Cellexperimentsshowedthattotalflavonoids fromsea buckthornbranchescould effctivelyreduce thedamageof ethanol to HepG2 cells,and protect HepG2 cells through the Keapl/Nrf2/HO-1 pathway.
