Analysis of Monosaccharide Composition of Tobacco Water-soluble Polysaccharides by Pre-column Derivatization HPLC

Abstract:Adeterminationmethod was established forthe monosaccharide compositionandcontentof tobaco water-soluble polysacharides by1-phenyl-3-methyl-5-imdazolinone(PMP)pre-columnderivatizationhigh performance liquidchromatography. Completeacidhydrolysis method was employedtohydrolyze tobacco water-solublepolysaccharides,andthe hydrolysis products weresubjectedtoPMPderivatizationreaction.Thesignalofmonosaccharidederivativesof tobacowater-solublepolysaccharides afteracid hydrolysis was determinedbyreversephase high-performance liquidchromatography.Themethod wasusedto explorethe monosaccharidecompositionandcontentof tobaccowater-solublepolysaccharidesofdiferentregionsandtypes.Theoptimized detection conditions were as follows: 0 . 0 5 m o l / L phosphate( -NaOH,pH7.O) buffer (solvent A) and acetonitrile (solvent B) were chosen as the mobile phase of elution system at the isocratic elution condition ( 8 4 : 1 6 ) ,with Agilent Eclipse XDB-C18 column ( 2 5 0 m m×4 . 6 m m, ; theUV detection wavelength was 2 5 0 n m .Results showed that the linear relationship is good for the nine monosaccharides in the concentration range of 2 0 . 0-8 0 0 μ g / m L ,with the correlation coefficient .The limits of detection and quantification were 0.07-0.17 and 0 . 2 2 -0 . 5 6 μ g / m L ,respectively. The average recoveriesranged 8 2 . 3 % - 1 0 4 . 0 % in spiked samples.Thetobacowater-solubleposaccharideswerecomposedofmanose (Man)hamose(Rha),galactose(Gal)aboe (Ara),glucose(Glc),glucuronicacid (GlcUA),and galacturonicacid (GalUA),with Glcand GalUA acounting forahigher proportion.There were significant differences inthe monosaccharidecompositionof tobaco water-soluble polysaccharides from variousregionsandtypes.Themethodisfast,simple,highlysensitive,andhasgoodrepeatability,thuscanbeused for monosaccharidecompositionanalysisof tobaccopolysaccarides.At thesametime,itprovidesabasisforfurtherresearchonthe structure of tobacco polysaccharides.
