Abstract:Anthracnose of Passflora edulis caused by Coletotrichum fructicola isa significant disease afecting the yield andqualityof pasionfruit.Elucidating thepathogenicmolecularmechansmsofColetotrichumfructicolaisof greatsignificance for disease control. In this study,we identified the small GTPase Rab7 from c . fructicola and compared its amino acid sequences withthoseof Rab7 proteins from various fungi.Phylogeneticanalysis revealed that theRab7proteinof c fructicola hadthemost distantrelationshipwiththeRab7proteinofScharomycescerevsiae,whileitwasloselyelatedtotheRab7proteinsofColeto trichumgloeosporioidesand Colletotrichumgraminicola.Using homologous recombination technology,we constructed the Rab7 gene knockout strain ΔRab7 and the complemented strain △Rab7/Rab7.Phenotypic analysis showed that comparedwith thewild type and Δ Rab7/Rab7,the Δ Rab7 mutantexhibited significantlyreducedcolonydiameter on PDAY and MM media,significantly increased melanin accumula

tion,and significantly decreased conidial production.However, Δ Rab7and the wild type N425did not show significant differences in sensitivity to H2O2 stress.Confocal laser scanning microscopy observations indicated that the Rab7-GFP fusion protein was localized to the vacuolar membrane,and the vacuoles in Δ Rab7appeared fragmented,suggesting that Rab7 gene maintained normal vacuolar morphology in c fructicola by regulating vacuolar fusion.This study provides a theoretical basis for elucidating the molecular mechanisms of pathogenicity in C* fructicola.
