法復(fù)制衰竭H9c2細(xì)胞模型。將細(xì)胞分為空白組,模型組,6-姜酚組, ,5,10,20μmol/L 去甲烏藥堿單用組以及分別與 20μmol/L 6-姜酚配伍組,共9組,觀察細(xì)胞的線粒體形態(tài),檢測線粒體膜電位、 Ca<sup>2+</sup> 濃度、ATP含量及線粒體膜部分主動轉(zhuǎn)運(yùn)酶活性。結(jié)果衰竭H9c2細(xì)胞復(fù)制條件為<img src="/qkimages/hnzy/hnzy202504/hnzy20250409-2-l.jpg" with="140px" style="vertical-align: middle;"> 干預(yù) 2h 與模型組相比,去甲烏藥堿單用及與6-姜酚合用均明顯改善模型細(xì)胞線粒體的微觀結(jié)構(gòu)損害。與單用去甲烏藥堿比較,配伍6-姜酚對模型細(xì)胞具有協(xié)同保護(hù)作用,模型細(xì)胞的線粒體膜更為完整、邊界清晰、嵴結(jié)構(gòu)恢復(fù),線粒體膜電位顯著升高,細(xì)胞質(zhì)及線粒體鈣濃度均大幅降低,同時(shí),ATP含量及 Na<sup>+</sup>-K<sup>+</sup>-ATP 酶 ?Ca<sup>2+</sup>-ATP 酶 Ω<sub>Λ</sub>Ca<sup>2+</sup>-Mg<sup>2+</sup>-ATP 酶活性均顯著增強(qiáng)( P< 0.05,P<0.01 )。結(jié)論去甲烏藥堿與6-姜酚配伍可顯著增強(qiáng)其對衰竭 H9c2 心肌細(xì)胞的保護(hù)作用,該作用可能與兩者合用協(xié)同緩解線粒體鈣超載、提高線粒體膜ATP酶活性、增加ATP的產(chǎn)量及促進(jìn)線粒體功能恢復(fù)有關(guān)。-龍?jiān)雌诳W(wǎng)" />

[Abstract]ObjectiveTocomparetheefectsofhigenamine beforeandaftercombinationwith6-gingerolon failing cardiomyocytes.Methods A failing H9c2 cell model was established using hydrogen peroxide ( H2(ρ)2) .The cells were divided into nine groups: blank group,model group, 6-gingerol group,higenamine single-use groups (5,10,20 μmol/l ),and higenamine combined with 6-gingerol (20 μmol/L )groups.The mitochondrial morphology of the cellswas observed,and the mitochondrial membrane potential, Ca2+ concentration,ATP content,and mitochondrial membrane-associated active transport enzyme activity were measured. Results The replication conditions for inducing failing H9c2 cells were determined as 1000 μ mol/L H2O2 for2 hours.Comparedwiththemodelgroup,bothhigenaminemonotherapyanditscombinationwith6-gingerolsignificantly aleviatedmitochondrialulrastructuraldamage.Comparedwiththeuseof higenaminealone,thecombinationwith6-gingerol hadasynergisticprotectiveefectonthemodelcells,asevidencedbyamoreintactmitochondrialmembrane,clearer boundaries,andrestoredcristaestructure.Additionall,mitochondrialmembranepotentialwassignificantlyhigher,cytoplasmic and mitochondrial Ca2+concentrations were markedly lower,andATP content,along withtheactivities of Na+- K+. -ATPase, Ca 2+, -ATPase,and Ca 2+, -Mg2+-ATPase, significantly enhanced ( P<0.05 , P<0.01 ). Conclusion The combination of higenamine and 6- gingerolcansignificantlyenhancetheprotectiveefectonfaiing H9c2cardiomyocytes.Thisefectmayberelatedtotheir synergisticactioninalleviatingmitochondrialcalciumoverload,increasingmitochondrialATPaseactivity,promotingATP production,and restoring mitochondrial function.
