AbstractObjectiveTo preliminarily explore the pro-angiogenic activity and mechanism of action of lignicol, based onthe zebrafish vascular injury model.MethodsVatalanib (PTK787) was used to incubate zebrafish embryos (24hpf) to construct a vascular injury zebrafish model,and they were exposed to lignicol atconcentrations of 5,10, and for 24h .Ferulic acid was used as a positive control to observe and calculate the vascular growth of zebrafish intersegmental and subintestinalvein vessels.The transcription levels of genes related to vascular growth in each group of zebrafish were determined using RT-PCR technology. ResultsCompared with the PTK787 model group,both the ferulic acid positive control group and Lignicol sample groups showed an increase in the length and numberof intersegmental vessls,as wellas the lengthof the subintestinal vein in zebrafish.Compared with the model group,lignicol (5,10, and )significantly increased themRNA expressionlevelsof related genesinthevascular endothelial growth factor pathway,includingvegf,vegfa,vegfc,vegfR2,erk2,akt,raf,pkband plcY. ConclusionThis study explores the pro-angiogenic effect and mechanism of compound lignicol through a Vascular injury zebrafish model for the first time.It was found that lignicolhas significant pro-angiogenic activityat concentrations of 5,10, and . The mechanism might raise the transcription levels of related genes in the VEGF signaling pathway,promote the proliferation and migration of endothelial cels,and thus increase the length and number of blood vessels.This study laid the foundation for the application development ofcompound lignicol.
