Abstract:Inordertorealizeindustrialproductionandaceleratetheopularizationandapplicationprocess,itisthebstwayto establish the plant tissue culture system of Clematis huchouensis.In this study,two-year old stem segments of clematis from Xing Wu with buds wereusedas explants fortissue cultureandrapid propagation.Theresults showed thatthe mixedtreatmentof 70% (20號 alcohol sterilization for 1O sand 10% sodium hypochlorite sterilization for1O min was the best,with a germination rate of 70.67% and a pollution rate of 17.33% .1/2MS medium is more suitable for start-up culture,and the sprouts grow well,not only robust,but also without yellowing and browning.In the multiplication culture, MS+2mg?L-16-BA+0.3mg?L-1 IBA +2 , L-1 hydrolyzed casein was the best,and the multiplication coeffcient was 4.73.The best roting culture medium was 1/2 ms+0.5 mg·L -1 IBA+0.01 mg·L-1 NAA,and the rooting rate was 79.67% .This study established a complete tissue culture technology system for Clematis huchouensis, breaking itsgegapicalitatios.Itprovidesathoreticalbasisforitspromotionandommecialdvelopent,ndlsoerse tain support for its interdisciplinary cooperation in the later stage.
