Establishment of Rapid Multiplication System for Tissue Culture of Cornus hongkongensis subsp.elegans RUAN Shuming,LAI Shigan, ZHOU Xianzhen

Abstract:InordertoestablisharapidpropagationsystemforCornusflorida,tisseculturemethodswereused.Thestudyinvolved screenigsterilizationmethdsforstemsegmentexplantsofCorusflordandinvestigatingtheefectsofdiferentconcentratinsof hormonesNAAandBAoallsiuction,uddfrentiatio,oungleafproliferationdotigTeresultsidiatedtste ilization with 0.1% HgCl2 for 12 minuteswas optimal.Callus induction was most effectivewith M IS+1.0mg?L-16-BA+0.2mg?L-1 NAA, reaching a 90% induction rate after 40 days.Bud differentiation from callus was best promoted by MS+2.O mg·L -1 6-BA+0.2 mg·L-1 NAA. Shoot proliferation from sterile young leaves was highest with MS+1.O mg·L-1 NAA+0.5 mg·L-16-BA.Root induction was best in MS+0.5 mg·L-1 IBA,however,the induced roots were prone to browning,necessitating timely subculture onto a rejuvenationmediumIncondusion,thisstudyprovidesatechnicalfoundtionforthelarge-scalepropagatinofthisspecies,whichisof great significance for its resource development and industrial application.
