Effect of chromium stress on the physiological characteristics of apple tissue culture seedlings

,China;3.AppleTechnology InnovationCenterofShandongProvince,Tai'an,Shandong,China) Abstract: The effects of different concentrations of potassium dichromate on the physiological characteristics of P1 tissue cultured seedlings of apple rootstock were studied.The subculture medium was treated with potassium dichromate at four concentrations of25,50,100 and 200mgL with 0mg/L asthe control.Physiological indicators were measured every 7 days after treatment.The results showed that tissue cultured seedlings were negatively affected by 25~200mg/L chromium treatment,which was positively correlated with the treatment concentration and varied with the extension of treatment time until it decreased.The chlorophyllcontent in seedlings decreases with
increasing treatment concentration;After the same treatment, the content began to decrease on the 21st day,and the greater the chromium treatment concentration,the greater the decrease; The activities of soluble sugars,antioxidant enzymes SOD,POD and CAT increased with the increase of chromium treatment concentration,and showed a trend of first increasing and then decreasing with the prolongation ofchromium treatment time.After 7days of treatment,low concentrations of 25and 50mg/L potassium dichromate had little effect on the seedlings,and the enhancement of antioxidant enzymeactivity was relatively small.At14or21 daysof treatment,the activities of antioxidant enzymes SOD,POD and CAT in the seedlings treated with high concentrations of 100 and 200mg/L decreased,and the chlorophyll content decreased.The seedlings were subjected to chromium stress toxicity.
