馬鈴薯無糖組織培養(yǎng)的條件優(yōu)化及其在馬鈴薯原原種生產(chǎn)中的應(yīng)用

Abstract:Tofurtheroptimizethemicroenvironmentparametersofsugar-free tisseculture,this studyused Qingshu No.9 as the tst material and designed different gradient experiments with diferentratiosof vermiculite to nutrient solution ( Vvermiculite : 1 Vnutrientsolution=1.0:0.7,Vvemiculie:Vnutrientsolution=1.0:0.8 mtriensoution =1.0:0.9),COconcentrtions(500 μmol/mol, 800μmol/mol , 1100μmol/mol , 1 400μmol/mol ),and light intensities to investigate the effectsof each treatment
onthe growth and development of potato tissue-cultured seedlings.Thelight intensitywasset at 50μmol/(m2?s) 75μmol/(m2?s) , 100μmol/(μ2?s). The results showed that the optimal conditions for sugar-free tissue culture of potatoes were as follows : , CO2 concentration of 800μmol/mol ,and light intensityof75 μmol/(m2?s). Compared with traditional tissue culture,the yield,quantity,and survival rate of potato pre-basic seeds obtained through sugar-free tissue culture were significantly increased by 16.09% , 22.37% ,and 3.27% ,respectively( P<
0.O5).Inconclusion,thisstudynotonlyclarifidthekeytechnicalparametersofsugar-freetissueculturebutalsoonfimedthe advantagesoftistechologyinimprovingteproductionefiencyofpotatoseedtubers,providingteoreticalbasisadtecal support for the standardized production and industrial propagationof potato tissue-cultured seedlings.

植物無糖組織培養(yǎng)技術(shù)由日本千葉大學(xué)古在豐樹教授發(fā)明[1],其技術(shù)核心在于通過精確控制微環(huán)境因素(如 CO2 濃度、溫度、濕度等),以環(huán)境 CO2 替代培養(yǎng)基中添加的外源糖作為碳源,實(shí)現(xiàn)植物完全自養(yǎng)生長(zhǎng)。(剩余10515字)
