Optimization of Extraction Process of Protein from Coprinus comatus and Study on Its Antioxidant Activity

Abstract: In order to determine the optimal extraction process of protein from Coprinus comatus by alkali extraction and acid precipitation method and analyze the antioxidant activity of Coprinus comatus protein,in this paper,with defatted Coprinus comatus as the raw material and protein extraction rate as the index,the effects of liquid-solid ratio,alkali extraction time,alkali extraction temperature and pH on the extraction rate of Coprinus comatus protein are studied. With ferrous ion chelation capacity, DPPH free radical scavenging capacity and hydroxyl free radical scavenging capacity as the indexes,the antioxidant activity of Coprinus comatus protein is evaluated. The results show that the optimal extraction process is liquid-solid ratio of 22:1 ,alkali extraction time of 126min , alkali extraction temperature of 48°C and pH of 12. At this time,the extraction rate of Coprinus comatus protein is 18.49% Within the mass concentration range of 2~10mg/mL ,the ferrous ion chelation capacity,DPPH free radical scavenging capacity and hydroxyl free radical scavenging capacity of Coprinus comatus protein increase with the increase of mass concentration,and the antioxidant capacity also increases,indicating that Coprinus comatus protein has certain antioxidant activity. This study has provided a certain reference basis for the further development and utilization of Coprinus comatus resources.
