Abstract: ObjectiveTo screen the optimal ratio of the PVA/BSP hydrogel matrix to prepare rifampicin loaded ROS-responsive hydrogel,improve the bioavailabilityofrifampicin,evaluate itsROS-responsive drug release and antibacterial efect,and provide a new optionfor bacterial infected wounds.MethodsTheappropriateconcentration ratio of PVA/BSP hydrogel matrix was screenedand optimized,and the ROS-responsive hydrogel was prepared by loadingrifampicin.TheROS-responsive drug release characteristicsof the hydrogel were measured by ultraviolet spectrophotometer,and its antibacterial property was further evaluated in vitro.ResultsThe optimal PVA/BSP ratio was 2:3 ( 8%PVA , 12%BSP ).The adhesion strength of the rifampicin-loaded hydrogel was 56.83KPa at a 15 mm stretch distance,and the highest compressive strength was 96.42 781 KPa.Compared with the PBS-released group, the rifampicin hydrogel showed a higher cumulative drug release in the H2O2 -PBS solution,and the rifampicin release rate at 48h (92.86%) )Was significantly higher than that in the PBS-released group (47.53%) .Invitro antibacterialtestresultsshowed thatthe inhibition Zoneof the hydrogel to methicilin-resistant Staphylococcus aureus, Staphylococcus aureus, and Escherichia coli was 3.4±0.10cm , 1.75±0.05 cm and 1.8±0.01cm ,respectively. ConclusionRifampicinhydrogel has goodbiodegradability,tisueadhesion,mechanical properties,ROS-responsive drug release ability,and goodantibacterial properties.Therefore,rifampicin hydrogel has a broad applicationprospect in the antibacterial and repair of bacterial infection wounds.
