Mechanism of Action of PLIN5 in Improving Ventricular Function in Pulmonary Arterial Hypertension by Regulating Fatty Acid Metabolism

【Abstract】ObjectiveToinvestigatetheoleandpotentialinfluenceofperilipin5（PLI5）intheabormalitisoflipidmetabolism inhypoxicpulmonaryhypertensionrats.MethodsTheratmodelsof hypoxicpulmonaryhypertensionwereconstructedbymeansofa hypobaricoxygenchamberandrightventriclarstructureandfunctionandhaemodynamicindexesreasessedinoal/Pkockout miceinanormal/hypoxicenvironmentforevaluatingrightventricularfunction.TheefectsofhypoxiaandPLN5onfatyacidmetabolism wereaessedbysystemiclipidmetabolismcumulationandchangesinlocalfattcidmetabolitelevelsinteartTeectsofyoia andPLIN5ontheexpresionandactivityofkeyenzyesinfattacidmetabolismweresubsequentlyevaluatedtoexploreteotential influenceofPIN5onfattacidmetabolism.Byevaluatingthestructureandfunctionofcardiomyocytemitochondria，weexploredthe protectivefectofI5onmitochondriathoughtheregulationofattyacidmetabolism.ResultsHypoxicpulmonaryhypertesionwas asociatedwithsructuralhangsndfuntioaldifteightveticleichereaggavatedy5eficiecytat acidmetabolisitsderoaloditiosutispathonic.Inatsioicpuoaryhertesiosigcatly fattacidetabasdencdbyeoagfttcidsisdteitatiooffatycidodatiogatcid synthesisndoxidationbymodulatingthactivitiesoffatyacidsynthaseandhormone-sensitivelipase.Meanwhile，LI5canprotect cardiomyocytemitochondriafrommitochondrialdamagecausedbyabnormalfattyacidmetaboismbyregulatingfattacidmetabolis.

ConclusionPLIN5protectshypoxicpulmonaryhypertensioncardiomyocytesfrommitochonrialdamageinducedbyabnomalfatyacid metabolism by regulating fatty acid metabolism of fatty acid synthesis and oxidation.
