Abstract:Fluorescence lifetime imaging is atechnique widelyused in various fields.Ithas high sensitivity,high timeresolutionandspatialresolution.Moreover,italsocan beused toquantitatively study thesamplewithoutthe influence of excited laser and environmental factors.Because of the above advantages,fluorescence lifetime imaging hadbeenwidelyused inbiomedicalfield.Fluorescencelifetimeimaging techniquecanbedivided into frequencydomain and time domain routines.Frequency domain routinecanbe divided into heterodyne method and homodyne methodaccording tothedifference frequency characteristics.There arethree major methods torealize time-domain routine,i.e.gating method,time-correlated single photon counting method(TCSPC)and scanning camera measurement method.This work introduces the basic principles of aforementioned fluorescence lifetime imaging methods. Wethenreview the recent application offluorescence lifetime imaging in biological sciencesandthe clinic study of tumorcancercell,and further give itsperspectives.
