[ABSTRACT]ObjectiveTo investigate the efect ofthephotothermal agent Cy7-TCFonoxidative stressand apoptosis in human pancreaticcancercels.MethodsMiaPaCa-2cellswere divided intocontrol group,Cy7-TCF group(treated with Cy7- TCF alone),and Cy7-TCF + laser group (treated with Cy7-TCF followed by 808nm laser irradiation). CCK-8 assay was used to measurecellviabilityunderdiferentconcentratiosofCy-TCFwithorwithoutlaseriradiation;fluorescecemicroscopywasused toobservetheuptakeofCy7-TCFbycels,andconfocal microscopycombined withalysosomalprobewas usedtoobserve thesubcelular localizationofCy7-TCF;the DCFH-DA fluorescent probewasused tomeasurethelevelof reactiveoxygenspecies(ROS) in cels;flowcytometrywasused tomeasurecellapoptosisrate.ResultsCCK-8assayshowedthatunderlaserirrdiatio,el viability significantly decreased with the increasing concentrationof Cy7-TCF in aconcentration-dependent manner( P<0.05 ). Fluorescence microscopyshowed that Cy7-TCF was efectivelytakenupby Mia PaCa-2cels,andconfocal microscopy showed that Cy7-TCF was mainly localized within lysosomes. DCFH-DA probe detection showed that under 808nm laser irradiation,Cy7-TCF significantly increased the level of ROS in Mia PaCa-2 cells ?q=19.07,16.84,P<0.05) . Flow cytometry showed that the Cy7- TCF+laser group had a significantly higher apoptosis rate than the control group and the Cy7-TCF group ( ?q=28.78,24.89,P< 0.05). ConclusionCy7-TCF is effectivelytakenupbyMiaPaCa-2celsandislocalizedinlysosomes,andthrough itsphotothermal effect,itinduces the increase inthe levelof ROS incells,reduces cellviability,and promotes cellapoptosis.
