Abstract:Thefungus Trichodermaspp.is ubiquitouslydetectableinsoil andplantrots,and it playsacrucial roleinthe biologicalcontrolof agriculture.To explore the influenceof Trichoderma spp.ontheresistanceof Astragalus membranaceus to rotrot,thisstudyisolatedandidentifiedTrichodermaspp.fromAstragalus membranaceusrootrot,andsubsequently investigated itsantagonisticactiononpathogenandanalyzed itsefectsonphysiological indexes of diseaseresistanceof Astragalus membranaceus,determined the typeof Trichodermaspp.throughmorphological characteristics,ITSand tef1 sequenceanalysis,analyzedantagonistic effects of Trichoderma spp.isolate on Fusarium solani HYFS-1 byplate confrontationassy.Thisstudyalso determined thephysiological indexessuch ascatalase(CAT),superoxidedismutase (SOD),phenylalanine ammonia-lyase(PAL),peroxidase(POD)and proline(Pro)to clarify the roleof Trichoderma spp.isolate ininducingthediseaseresistanceof Astragalusmembranaceus.Theresults wereasfollows:(1)TheTrichoderma spptype isolated from Astragalus membranaceus root rotwas identifiedas Trichoderma harzianumandnamed T9131.(2) Theinhibition rate of Trichoderma harzianum T9l31 against Fusarium solani HYFS-1 reached 72%±1% after6days of confrontation.(3)Compared to HYFS-1alone,T9131significantlyenhanced SODactivityat0hafter HYFS-1infection;at the 24 h HYFS-1 infection,T9131markedlyincreasedSODactivity,PODactivity,andProcontent;bythe48hHYFS-1 infection,T9131significantlyelevatedPODactivity,PALactivity,andProcontent.Tosumup,Trichodermaharziaum T9131 cancontrolAstragalus membranaceus rotrotbydirectlyinhibitingthe growthof HYFS-1andinducingtheactivities of SOD,POD,PALandProcontentof Astragalus membranaceus under Fusarium solani HFS-1 stres.Thisresearch will laythefoundationforexploring thebiocontroleffectofTrichodermaharzianumanditsmachanismofresistanceofAstragalus membranaceus root rot.
